Overview
Humanin was the first mitochondrial-derived peptide discovered and remains the most extensively studied member of this peptide class, originally identified in a screen for factors protecting neurons from Alzheimer’s-disease-related toxicity.
Mechanism & Research Findings
Humanin is studied for cytoprotective signaling through multiple pathways, including binding to the CHRNA7/WSX-1/gp130 receptor complex to activate STAT3 signaling, as well as direct interaction with pro-apoptotic Bax protein to inhibit mitochondrial-mediated apoptosis. Research has examined its neuroprotective activity in models of neurodegeneration, along with broader roles in cellular-stress resistance, positioning it as the foundational reference compound in mitochondrial-derived-peptide and cytoprotection research.
Research Applications
- STAT3 signaling and cytoprotection research
- Apoptosis (Bax pathway) inhibition studies
- Neurodegeneration and neuroprotection modeling
