Overview
FOX04-DRI is a cell-penetrating peptide engineered in a D-retro-inverso configuration (protecting it from rapid proteolytic degradation) that disrupts the interaction between FOXO4 and p53 specifically within senescent cells, a compound at the center of “senolytic” cellular-aging research.
Mechanism & Research Findings
In senescent cells, FOXO4 binds and sequesters p53 in the nucleus, blocking p53-driven apoptosis and allowing senescent cells to persist (“zombie cells”). FOX04-DRI competitively disrupts this FOXO4-p53 interaction, freeing p53 to trigger apoptosis selectively in senescent cells while sparing healthy, non-senescent tissue in published mouse models — a mechanism widely cited as a foundational proof-of-concept for peptide-based senolytic research.
Research Applications
- Cellular senescence and senolytic pathway research
- FOXO4-p53 protein-interaction studies
- Selective senescent-cell apoptosis modeling
